Mushroom spores require Mycelium in order to be able to grow. This can be obtained by using infestation spores on dirt. An infestation spore can be crafted by throwing any type of mushroom into your mana pool. This might seem like a Catch-22, in that you need mushrooms in order to grow mushrooms, but there are opportunities to obtain mushrooms before you get Mycelium. The Nether is one such source, if you manage to find a glowing mushroom from Natura and break it.
